Matlab Image Processing

I work for a Cancer Research Lab. My job is use Matlab for image Processing. I am given two images of a person's skin lesion, that has a tumor, at different wavelengths. The first image has the surgeon's markings on it (640 nm), which is the surgeon's estimate as to where the tumor is. The second image is a high frequency image of the tumor ( 440nm) and surrounding areas on the skin. My job is to make a composite image (using either the subtract function or divide function) to show the surgeon a better estimate of the size and location of the tumor in relation to the surgeon markings. The images will be in grayscale and then pseudocolor will be added at the end. I need someone to write a program for me that can take a set of images ( 2 images at different wavelength of the skin tumor) and enhance the images in a way that is specific to the image ( I think this may require computer vision) and make a composite image from the two images using either the subtraction or division of the two images. The hard part for me is to create a program that can do specific image enhancement for a given image ( contrast adjust, sharpen edges, make the image overall more clear to see for the surgeon). I wrote a basic function but my function does not work on any image that is inputted which is a problem. I have attached an iphone picture using another software called Fiji (imageJ) what it should roughly look like on matlab. The two images on the left are the pictures of the skin tumor lesion and the image on the right is the composite pseudocolor image. I will also attach two TIF images of the same skin lesion taken at different wavelengths so you can use as practice ( they might appear all black but they images will appear once you adjust the contrast). Please help me, if you want me clarity please ask. To be clear, I know how to manually do this in matlab, but I don't know how to write code that will take any two images and will enhance the images according that is specific to each individual image.
